Tanami, Yoh (University of Texas Medical Branch, Galveston) and Morris Pollard. Effect of p-fluorophenylalanine on psittacosis virus in tissue cultures. J. Bacteriol. 83:437-442. 1962.-The inhibitory effect of p-fluorophenylalanine (FPA) on maturation of psittacosis virus was investigated, with attention to the time sequence of viral protein synthesis. Extracellular virus particles were not inactivated by FPA at a concentration of 100 mug per ml, at which level it interfered with maturation of intracellular virus. When FPA was added to infected tissue cultures earlier than 15 hr after infection, intracellular virus maturation was suppressed. However, when FPA was added after 15 hr, infective virus was produced, which indicates that the synthesis of a FPA-sensitive virus precursor (presumably viral protein) had already occurred. A latent ("dormant") infection of psittacosis virus, established in a medium deficient in phenylalanine and tyrosine, was also investigated.

The effect of FUDR on vaceinia virus replication in Heba cells was investiga~d. The incorporation of deoxyuridine-Ha into viral DNA, as determined by autoradiography, was completely blocked by 10-e M FUDR.. This concentration of the analog prevented production of infect, ious virus above levels present in the cultures after washing at the end of the viral adsorption period. The production of vac...

The effect of the amino acid analog FPA on vaccinia virus replication in HeLa cells was investigated, By varying the dose of FPA or the time after infection at which the analog was added to cultures, it was possible to inhibit differentially the synthesis of viral components in vaccinia-infected cells. When suflicient FPA was added t,o GUItures at the t,ime of infection, the synthesis of viral ...

A complication in the analysis of the role of ICP34.5 gene in the herpes simplex virus type-1 (HSV-1) lifecycle is the presence of overlapping antisense gene, open reading frame P (ORF P), which is also deleted in HSV-1 ICP34.5 negative mutants. A HSV-1 wild type strain (17+) ICP34.5/ORF P deletion mutant (1716) is totally avirulent in animal models and impaired in a number of in vitro function...
متن کاملFactors Related to the Growth of Psittacosis Virus
Previous studies have shown that it is possible to interfere with the intracellular growth of psittacosis virus (6BC) in tissue cultures by the addition of analogues of certain metabolites (1-3). These earlier investigations have established the importance of pteroylglutamic acid (1), citrovomm factor (folinic acid) (1), adenine (2), guanine (2), and uracil (3) for the propagation of the virus ...
متن کاملFactors Related to the Growth of Psittacosis Virus (strain 6bc)
The inhibitory action of sodium sulfadiazine on the growth of psittacosis virus (6BC) in embryonated eggs is readily reversed by citrovorum factor but not by small amounts of vitamin B(12). In embryonated eggs, the pteroylglutamic acid analogues, 9-methylpteroylglutamic acid and 4-aminopteroylaspartic acid, produced some suppression of the growth of psittacosis virus (6BC). 4-Aminopteroylglutam...
